Structure-function relationship of alpha-synuclein fibrillar polymorphs derived from distinct synucleinopathies

Alpha-synuclein aggregation represents a hallmark of several neurodegenerative diseases, including Parkinson’s disease (PD), Multiple Systems Atrophy (MSA), and Dementia with Lewy bodies (DLB). In this talk, Dr. Tetiana Serdiuk will unveils the structural diversity of alpha-synuclein fibrillar polymorphs across these conditions.
Using cutting-edge Limited Proteolysis coupled with Mass Spectrometry (LiP-MS), her research demonstrated that alpha-synuclein fibrils amplified from patient brains are not only structurally distinct but also exhibit unique interactomes that affect different neuronal pathways upon internalization. Additionally, native alpha-synuclein structures in the brains of PD, DLB, and MSA patients were found to differ from one another. These structural differences are directly linked to unique fibril interactomes and cellular responses, providing new insights into disease mechanisms. Leveraging CRISPR-based tools, she has identified key proteins that modulate alpha-synuclein inclusions in a polymorph-specific manner—potentially unlocking new therapeutic targets for these untreatable diseases.
